چکیده
Abstract We have developed an innate immune model of colitis that occurs in the absence adaptive immunity, is early-onset, 100% penetrant, chronic, and dependent on microbes. villin-TNFAIP3 transgenic mice, crossed to RAG1-/- mice (aka TRAG mice) develop immune-mediated not observed or littermates. Antibiotic treatment prevents mice. Thus, we sought investigate microbes drive chronic immunity. To examine spatial organization bacteria, mucus intestinal epithelium, bacteria were localized Methyl-Carnoys-fixed sections WT, villin-TNFAIP3, colons layers collected by laser capture microdissection (LCM) for evaluation bacterial populations. test antibiotic effectiveness prevention, treated with single solutions ampicillin, neomycin, metronidazole, vancomycin, all antibiotics combined a cocktail, was scored along proximal-distal axis colon. Microbial populations gut compared between 16S sequencing. invasion layer abundant but WT LCM indicated subset microbes, classes Gammaproteobacteria Actinobacteria, over-represented These included increased abundance genera Serratia, Acinetobacter, Pseudomonas Corynebacterium decreased Bifidobacterium. Differences colon, neomycin being most effective prevention inflammation cecum, while ampicillin distal The communities distinct tightly grouped within proximal whereas this structure lost In gut, relative Firmicutes (mainly Lachnospiraceae Ruminococcacaea) Proteobacteria Helicobacteriaceae Desulfovibrionbacteriaceae) Actinobacteria Bifidobacteriaceae) Bacteroidetes Bacteroidales). Distinct Microbial Communities Trigger Colitis Development upon Intestinal Barrier Damage via Innate or Adaptive Immune Cells
Inflammatory bowel disease comprises a group of heterogeneous diseases characterized by chronic and relapsing mucosal inflammation. Alterations in microbiota composition have been proposed to contribute to disease development, but no uniform signatures have yet been identified. Here, we compare the ability of a diverse set of microbial communities to exacerbate intestinal inflammation after che...
متن کاملInnate immune detection of microbial nucleic acids.
Detection of pathogen-derived nucleic acids by pattern recognition receptors (PRRs) is essential for the host to mount an appropriate immune response, which for viruses involves the induction of type I interferons (IFNs). By contrast, inappropriate activation of PRRs by self nucleic acids can lead to autoimmunity. Recent developments in PRR research have uncovered important new molecular detail...
متن کاملImmune-complex mediated colitis in rabbits
An experimental colitis in rabbits is described, following the intravenous injection of preformed irnmune complexes of human serum albumin (HSA) and anti-HSA into non-sensitised rabbits. Tissue damage was localised to the colon by the Auer technique ofinducing local non-specific inflammation, by the rectal instillation of dilute formalin. Formalin alone gave transient changes that reverted to n...
متن کاملPersistently Active Microbial Molecules Prolong Innate Immune Tolerance In Vivo
Measures that bolster the resolution phase of infectious diseases may offer new opportunities for improving outcome. Here we show that inactivation of microbial lipopolysaccharides (LPS) can be required for animals to recover from the innate immune tolerance that follows exposure to Gram-negative bacteria. When wildtype mice are exposed to small parenteral doses of LPS or Gram-negative bacteria...
متن کاملHistiocytic Ulcerative Colitis: Infectious or Immune Mediated?
The development of culture independent techniques for determining the presence of infectious agents has revolutionized the approach to detecting both suspected and unsuspected pathogens. For example: PCR with broad range primers has led to the identifi cation of Bartonella henselae as the agent of cat scratch fever and peliosis hepatis. PCR with pathogen specifi c primers has enabled the accura...
